Impact of the ICHQ9: A Case Study, Filling Operation

Risk assessment approach is becoming more popular in pharmaceutical industry since FDA started talking about:

The Gold Sheet – May 2000 – Vol. 34, n°5:"Risk management is being explored by FDA as a possible next step in the evolution of pharmaceutical good manufacturing practice."

…"Risk management approach… is under consideration as a mechanism to advance drug GMP compliance”
FDA Strategic Action Plan – Aug 2003“Use science-based, efficient risk management in all agency regulatory activities, so that the agency’s limited resources can provide the most health promotion and protection at the least cost for the public.”

In line with this strategy, ICH issued in November 2005 ICHQ9, the Quality Risk Assessment Guideline with the purpose to offer a systematic approach to quality risk management and provide principles and examples of tools to be applied for different aspects of pharmaceutical quality.
The purpose of the guideline is not to solve the issues, but provide different tools that can be helpful in solving them. In particular the objective is to identified the right tool (a tool is the suggested technique to approach the risk analysis) for the specific scope (i.e. route cause of an investigation, identification of process critical parameters).
The Risk Assessment approach is a very powerful way to document the rationale associated to the risk exposure evaluation for an aseptic process.
As a case study, we will see how Risk Assessment can be successfully applied in identification of RPI (risk priority index) associated to the aseptic connections executed to assemble a filling line. The filling line investigated in the case study is located in grade A area (ISO5), fills in vial a broad range of sterile products: suspensions, ointment, solution not terminally sterilized (aseptic filling). The line doesn’t have a CIP and SIP system, so the machine set up represented a critical path for product contaminations due to the huge numbers of aseptic connections.

Impact of the California Supreme Court’s Recognition of the “Sophisticated User Defense” on Life Sciences

In Johnson v. American Standard, Inc., 2008 WL 878933 (Cal. Apr. 3, 2008), the California Supreme Court held that a manufacturer is not liable to a sophisticated user of its product for failing to warn of dangers about which the sophisticated user knew or should have known. The seminar will examine in more detail how various factors of weathering and climate, such as solar radiation, heat and moisture, may affect materials and how to test the resistance of a formulation or product to them. We will discuss how measuring devices for parameters of weathering function, and identify some of the common errors associated with their use. We will discuss in more detail the way advanced laboratory equipment controls irradiance, humidity, and temperature. Visual and instrumental evaluation methods will be discussed for both appearance and physical attributes. As with the original Fundamentals of Weathering course, special attention is given to the testing techniques of paints and protective coatings, automotive materials, architectural building products, molded plastics, and textile materials.
